Australia says 294 polluters to pay carbon tax

(AP) ? The government announced Friday that 294 of Australia's biggest polluters will pay a carbon tax when it comes into force next month in a bid to curb the nation's greenhouse gas emissions.

Opinion polls show that the carbon tax is already deeply unpopular among Australians. But Prime Minister Julia Gillard maintains it is essential to achieving her 2020 target of slashing greenhouses gas emissions to 5 percent below 2000 levels.

Companies and local governments that annually produce more than 25,000 metric tons (28,000 tons) of carbon dioxide, or the equivalent in other carbon gases, must pay a tax of 23 Australian dollars ($23) for every metric ton of carbon dioxide they produce starting July 1.

BHP Billiton, the world's biggest miner, and Macquarie Generation, one of Australia's largest power generators, are on the list of carbon taxpayers, as are local governments that operate methane-emitting landfills.

The agricultural industry, Australia's third worst carbon polluter, is exempt because of difficulties in measuring emissions on farms.

Climate Change Minister Greg Combet said the number of carbon taxpayers could change in the months ahead as businesses either increase or decrease emissions.

Australia is one of the world's worst greenhouse polluters on a per capita basis due to its heavy reliance on abundant reserves of cheap coal for electricity generation.

(AP) ? He's come out of the shadows and into the world spotlight. But Massimiliano Giornetti, Salvatore Ferragamo's creative director since 2010, says that he's still a "down to earth" guy ? despite having tea with Meryl Streep and hanging out with Angelina Jolie.

The humility is perhaps surprising considering that Giornetti, who has breathed life into the 84-year-old fashion house, made history this week with the biggest catwalk coup in recent times: taking over the arcades of the Louvre for the 900-year-old building's first fashion show.

While the Louvre venue might be new, the Hollywood A-list has been linked for an eternity to the storied Italian brand.

"The link between Salvatore Ferragamo and Hollywood is as old as the company," Giornetti told The Associated Press. "It was probably the first house (in the 1920's) to create this link between fashion and celebrities."

It was patronage of actresses ? such as Marilyn Monroe and Greta Garbo ? that ensured Salvatore Ferragamo's "fairytale" rise from a humble shoe repair shop in California into the leather "shoemaker of the stars."

The ongoing Marilyn Monroe exhibit in Florence's Museo Salvatore Ferragamo is proof enough of the brand's star quality. It features 14 pairs of the house's shoes once worn or purchased by the screen siren.

Today, Ferragamo is one of the world's biggest fashion empires? spanning leather goods, watches, perfumes and ready-to-wear for men and women. Sales last year were $1.37 billion.

Giornetti says Ferragamo's a natural hit because of a simple and universal concept: beauty with functionality.

"The first time Greta Garbo visited the Ferragamo headquarters," Giornetti recounted, "the diva said 'I want to have shoes to walk with.'"

That eye for comfort, stamped on the house from the beginning, was what made the shoes so famous. Salvatore Ferragamo ? the founder of the house who died in 1960 ? even studied human anatomy to master how to make elegant shoes truly wearable.

From this know-how sprang the famous wedge heel in 1936. Then later came the classic staple bestseller ? the Vara Bow Pump.

Ferragamo ensured this philosophy was carried on to Giornetti, half a century later.

A native of Tuscany, Giornetti studied in Florence. After 10 years designing Ferragamo's menswear, Giornetti was named creative director in 2010, giving him control of the women's side, too.

With Ferragamo's headquarters situated in Florence's Palazzo Spini Feroni ? a fortified medieval edifice ? it's clear that both the designer and the brand have the city in their hearts.

Florence's hands-on artisanal tradition has existed since the Renaissance. Giornetti says it's key to the house and to his own personal style, mixing aesthetics with practicality.

"It's modern teaching that started back in the past. (Ferragamo's philosophy) is linked with the work of Leonardo da Vinci, who also studied the anatomy and the human figure."

It's thanks to Ferragamo's links and sponsorship of da Vinci's restored masterpiece "The Virgin and Saint Anne," that the Louvre granted access to Tuesday's show ? a showcase of clothes inspired by Florence's artisanal history.

In the collection, relaxed cool-colored clothes with intricate and detailed stitching were combined appealingly with Florentine leather savoir-faire.

There was a nod to couture craftsmanship in a knee-length cream dress with myriad organic looking leather appliques.

Though he just showed in one of the world's most famous buildings, Giornetti remains unpretentious.

He lives in the non-trendy part of Florence on the left bank because "it's the most authentic and genuine part of the city, where you find the artisanal traditions: the goldsmith, the silversmith, the art of dyeing leather, the old tradition of basket weaving."

Given his roots it's no surprise that Giornetti praises the slower and less showy side to fashion. It's rooted in a simple, artisanal approach to clothes ? devoid of ego.

The down-to-earth values of a family-run label also appeal to him.

"Working for a family business means a lot, especially when everything is becoming more global with fashion brands becoming part of enormous companies."

Despite Giornetti's new-found fame ? people now want to take photos with him in the street ? he says he's still a "very down to earth person."

Yes, he's now able to rub shoulders with his idols like actress Angelina Jolie and Chinese film director Wong Kar-wai, but he says, "I still like to go to the cinema by myself."

It's with genuine pride that he speaks of a fitting he did for actress Meryl Streep in London ahead of last year's Academy Awards.

His favorite moment with the Oscar-winner? As it rained, "sharing a cup of tea."

BoE, UK Treasury to ease tight credit: Osborne

LONDON (Reuters) - Britain's government and the Bank of England will act together with new monetary policy tools to tackle tightening credit and financial market conditions triggered by the euro zone crisis, finance minister George Osborne said on Thursday.

Osborne said BoE Governor Mervyn King would reveal more details of the scheme, designed to protect Britain's ailing economy from the shockwaves of Europe's banking and sovereign debt problems, in a speech at 1600 EDT.

"The immediate priority is to counter the tightening of financial conditions and increase in bank funding costs caused by the crisis in the euro zone," Osborne told London's financial elite in a speech.

"I can tell you today that the governor and I will take coordinated action on liquidity and on funding for new bank lending in order to inject new confidence into our financial system and support the flow of credit to where it is needed in the real economy."

Osborne said he would also give details in the next few weeks of a scheme to boost private sector investment in infrastructure - a measure he says is possible due to Britain's strict austerity plan.

Britain's Conservative-led coalition government has leant heavily on the BoE to support the economy as ministers slash spending to reduce a record budget deficit.

Osborne said the BoE's Financial Policy Committee, a new body charged with safeguarding Britain's financial stability, would be required by law to support the government's economic policy goals.

"I will make it a legal requirement for the FPC to report, for every action it takes, how that action is compatible with economic growth as well as stability," he said.

SAN FRANCISCO, June 14 (Reuters) - The electricity was back as Tiger Woods moved into once familiar territory on Thursday, clawing his way into contention with a one-under-par 69 in tough scoring conditions to lie three shots off the U.S. Open lead.

As the year's second major lived up to its reputation as the toughest championship of all, the former world number one mixed three birdies with two bogeys to finish the first round three behind fellow American Michael Thompson at the Olympic Club.

"I played well today," three-times champion Woods told reporters after breaking 70 in the opening round of a U.S. Open for the first time since 2002 with a superb display of course management on a firm and fast-running layout.

"I felt like I had control of my game all day. I'm really excited how I was able to execute my game plan all day today. The golf course was really quick.

"We knew the greens were going to be a little quicker, but I didn't think they would be this firm this early in the week. So we had to make a couple of adjustments with that."

Woods was one of just six players who broke par, ending the round tied for second with fellow Americans Nick Watney and David Toms, Britain's Justin Rose and 2010 champion Graeme McDowell of Northern Ireland.

Watney capped a memorable display in glorious afternoon sunshine with a rare albatross two at the par-five 17th, holing out with a five-iron from the fairway for only the third double-eagle ever recorded at a U.S. Open.

The little known Thompson, who tied for 29th in his only previous U.S. Open appearance in 2008, upstaged the game's biggest names by carding a seven-birdie 66 on the challenging, hilly Lake Course.

Though Thompson bogeyed three of the first six holes - a stretch widely regarded as the most difficult start in the majors - he then surged up the leaderboard with six birdies in bright sunshine to take control of the tournament.

"This is one of my favourite golf courses, so I've got good feelings coming in here," said the 27-year-old Thompson, who booked his place in this week's field via sectional qualifying.

"I just got the putter hot today. I got kind of a wake-up call on the first hole, I missed my par putt because I didn't realise the greens were that fast. From then on, I was just trying to flow."

American world number six Matt Kuchar, 2003 U.S. Open champion Jim Furyk, Swede Robert Karlsson and Britain's Ian Poulter were among a group of eight locked on 70.

Most of the players struggled on the tight fairways and surprisingly quick greens, Phil Mickelson carding a 76, defending champion Rory McIlroy a 77, Masters champion Bubba Watson a 78 and world number one Luke Donald a nine-bogey 79.

'TOUGH DAY'

"I didn't play very well," Mickelson said after a round including seven bogeys, one birdie and a lost ball with his opening tee shot, a wild hook into trees. "It was a tough day when you play it the way I did."

McIlroy, who romped to victory by eight shots in last year's U.S. Open at Congressional, paid the price for hitting only seven of 14 fairways.

"It's just so tough here if you put yourself out of position at all," the 23-year-old said. "When you're trying to play catch-up on this golf course it's very hard.

"I was able to make only one birdie out there today. I need to try to make more tomorrow and limit the mistakes."

Left-hander Watson never recovered from four bogeys in his first eight holes but was hugely impressed by Woods.

"That was the old Tiger," he gushed. "That was beautiful to watch. That's what we all want to watch and that was awesome to see him strike the ball look. He made a couple (of) bogeys but under par on this golf course is pretty good."

Woods, who won the most recent of his 14 major titles at the 2008 U.S. Open, oozed confidence throughout his round as he outshone playing partners Mickelson and Watson in front of huge galleries.

Now we're wondering how many people actually have an active and untreated phobia."

The abstract "Are people with insomnia afraid of the dark? A pilot study" from Ryerson University Sleep & Depression Lab is being presented today at SLEEP 2012, the 26th annual meeting of the Associated Professional Sleep Societies (APSS) in Boston.

Colleen Carney, PhD, the principal investigator, said insomnia treatments are highly effective but not everyone responds or completely recovers. New approaches may be warranted. For example, the most effective insomnia treatments encourage people to leave the dark bedroom and go into another, lit room; however, this would not be a way to treat a dark-related phobia.

"We may need to add treatment components for these patients and adapt existing treatment components in light of the phobia," Carney said. "A lot more research is needed, but we believe we have stumbled across an unmet treatment need for some poor sleepers."

(AP) ? The affable man with the horse that may become the first Triple Crown winner in more than a generation can't seem to outrun his unflattering nickname: "Drug" O'Neill.

But Doug O'Neill is far from the only trainer in Saturday's Belmont Stakes with a history of improperly medicated horses. The Associated Press reviewed the histories of all 11 trainers with horses in the race and found that 10 had at least one violation of medication regulations set by state racing boards.

O'Neill has been under the most scrutiny because his colt, I'll Have Another, won the Kentucky Derby and Preakness Stakes and is the 4-5 favorite to add the Belmont and complete the first Triple Crown in 34 years.

"We had the black cloud before he won the Derby," D. Wayne Lukas, the elder statesman among trainers, said of horse racing's drug problems. "Now it's just gotten darker."

Lukas, who will run 20-1 Optimizer in the Belmont, didn't mention his own record. He has had almost as many violations as O'Neill, though spanning a longer career with a larger stable and including none in the last 13 years.

That's something Penny Chenery, the doyenne of the sport and owner of the great Triple Crown champion Secretariat, apparently didn't realize when she told The Atlantic magazine that I'll Have Another's owner, J. Paul Reddam,"should be embarrassed that the trainer he has chosen does not have a clean record."

In fact, only one trainer in the Belmont has a clean record ? Kelly Breen, whose horse, My Adonis, was a last-minute entry. Five of the others have had a single violation, typically for medications commonly used either to control inflammation or to prevent internal bleeding while racing. Their use is legal only within bounds.

AP's review included hundreds of rulings from state racing commissions collected by the Association of Racing Commissioners International, which represents the sport's regulators. The majority of violations were unrelated to medications; improper paperwork was common, and there were a few for profane tirades as well.

O'Neill shrugs off his nickname and denies the behavior it implies.

"Not good," he said when asked how it makes him feel. "But it just happens that my name rhymes with that.

"You can say whatever you want. I know at the end of the day I love my horses and I take great care of my horses."

For the 11 Belmont trainers, AP found 64 medication violations in the association's database, which is regarded as the industry's most comprehensive. The database did not include two violations O'Neill had in California for elevated levels of carbon dioxide in his horses' blood. Adding those two, O'Neill had 17 rulings against him dating to 1997.

Only the two biggest names in the sport, Lukas and Bob Baffert, were anywhere close to that number. According to the association's data, Baffert actually had more, with 20, and Lukas had 15.

Dale Romans, who will saddle second-choice Dullahan, had five violations, four of them for improper administration of commonly used medications. His most recent were two violations three years ago in Florida. Ken McPeek, the only trainer with two horses in Saturday's race, had four violations, the last a positive test for the anti-inflammatory drug diclofenac in Illinois in 2005.

The five trainers with one violation were Manuel Azpurua, Chad Brown, Michael Matz, Dominick Schettino and Doodnauth Shivmangal.

One longtime testing official who reviewed O'Neill's violations record for AP said he didn't find it particularly egregious.

"There are a lot of people in racing that have records similar to his," said Richard Sams, director of the HFL Sport Science Laboratory, the official testing lab for Kentucky and Virginia's racing commissions. "He's getting a lot of attention right now obviously because he has the horse to beat."

The amounts that state boards fined Baffert and Lukas were much lower than O'Neill's total, generally reflecting the more routine nature of most of the violations. Lukas was assessed $500, Baffert $5,800 ? and O'Neill $32,550.

Lukas did, however, have one of the most serious violations ? a positive test for the narcotic painkiller oxymorphone ? more than 30 years ago. And Baffert ? who trains Derby and Preakness runner-up Bodemeister but will run 8-1 shot Paynter on Saturday ? got in trouble in 2001 after one of his horses tested positive for morphine. He blamed contaminated feed.

Baffert said the case was dismissed.

"If a trainer has a big barn, things are going to mess up," he said. "It's mainly mistakes."

While Lukas was last cited in 1999, and Baffert's horses have been relatively problem free in recent years, O'Neill's violations have been stacking up.

His latest troubles are particularly ill timed.

Just last month, the California Horse Racing Board decided to suspend him 45 days, starting after the Belmont, because one of his horses had elevated carbon dioxide levels in its blood. High carbon dioxide levels reflect a change in blood chemistry that is believed to help a horse combat fatigue by limiting lactic acid buildup.

While California's board didn't rule that O'Neill intentionally doctored the levels ? typically done by feeding the horse a "milkshake" of bicarbonate of soda, sugar and electrolytes ? the authorities concluded that because O'Neill is responsible for the care of his horses, he should be punished. Along with the suspension, he was fined $15,000. The horse finished eighth.

O'Neill vigorously fought the most recent charges, and still can appeal. It was the third time California's board sanctioned him for an elevated carbon dioxide level in the past several years, to go with one in Illinois in 2010.

This year, New York state racing regulators reinstated a rule that horses in the Belmont be housed in a "detention barn" where their diets and medicines are strictly monitored. The explanation: They want to ensure that the race is run with clean horses.

Some trainers worry the change of scenery will upset their horses, and have bristled at the rule. Not O'Neill, whose comments have been, as usual, public-image savvy.

"I like the thought of showing the general public that all the horses are in the same locker room," he told reporters. "The transparency that our game probably lacks is key."

O'Neill has said all of his violations were for "therapeutic medications" in excess of allowable limits, not for banned drugs.

That is debatable, according to Sams, the testing lab official.

"I think from his point of view, he sees everything as something to help the horse ? and in his mind that is therapeutic," Sams said. "I think racing regulators see things a bit differently than he sees them, and with reason."

Sams specifically cited findings of the anti-inflammatory drugs etodolac and naproxen in O'Neill-trained horses as examples of drugs that are generally prohibited in the sport.

Not everyone was quick to indict.

"I don't think that anything Doug has done is on purpose, and if it's happened it's probably been for some silly reason," McPeek said.

Lukas called O'Neill a "good horseman" who wouldn't do anything illegal but said the problem is the perception "that horse wasn't perfect in the first two legs."

"That is a ridiculous assumption," he said. "Just that perception will be a black eye. People will say, 'Well, they've cleaned it up and now he got beat.' That's a terrible assumption."

Networking Interview #16: How To Create a Compelling Resume ...

Today in my networking challenge I am interviewing Karleen Harp. Karleen is a Professional Resume Writer and is better known as the Resume Smith. She specializes in helping early to mid-career professionals cross the bridge between their now and their future. And I am so excited to get all of her resume tips today, thanks so much for being here Karleen!

In case you are reading this at work and can?t watch the video, you can read all of the great advice below instead!

1) How can we make our resumes unique instead of boring and cookie cutter?

First, never use template that comes from your word processor. You want a clean and unique design that will draw the reader?s eyes to the key points that you want to demonstrate. The most important part of your resume is the top-half of page 1. So make sure your top selling points are up there. Second, know what your personal brand is and you need to sell it. Do this by describing not just what your previous job responsibilities were but it?s how well you did at it.? And the way that you do that is specific accomplishment statement. Many job seekers have a hard time remembering and don?t know thir business impact. A good resource is to go back to your previous performance review. And that?s how you become different. You need to show things that no else can claim because you did them. And that?s your accomplishment!

2) How many pages?

A lot of people say one page but that?s not necessarily true. The key is you don?t have unnecessary information in your resume.

3) Do you recommend using different resume for each job you apply to?

And it depends if you are applying for job in different job category. Make sure you are focusing your resume on what?s really important on that particular job.

4) What are your tips for cover letters? ?Do they even get read?

That?s an interesting dilemma. The truth is they don?t always read but you need to do it anyway. The reason why you need to do it is because some of them get read. And if you are applying to a company where the hiring manager or the recruiter really wants that information about you, you don?t want to roll yourself out because you are lazy. It is your why; why did I choose this career, why do I want to work to your company and why should you hire me. That?s the important thing that a cover letter should cover.

5) What makes a candidate standout from the other?

They have the ability to articulate what they can do for the company. A resume should be easy to read. Most of the recruiters right now are using applicant tracking system so they?re not looking at every resume they?re going to be using keyword searches instead. Keywords are really important if you are applying on an ATS.?My recommendation for job seeker is to avoid applying online at all costs and use networking to get a personal referral instead. Even if they say you need to apply to ATS, someone is going pulling your information out of the ATS if you have that personal referral to the company.

6) How can we use keywords in our LinkedIn profiles to get in more recruiter?s search results? ?Any other LinkedIn tips for job searchers?

LinkedIn is a large networking room. It just so happens that it is located in your computer instead of a conference center or something. When you go to a networking room, you are not going to only talk to people that you already know. You?re going to be walking up to people giving them a handshake and saying, ?Hi, I?m Anna! Tell me about yourself?. The same rule applies to LinkedIn. You want to purchase a page and group discussion, answer questions to show your knowledge and subject expertise for your target job. Having those conversations in those places can give you that handshake then send someone a connection.

7) So is it okay that our LinkedIn profile is a lot longer than our resume would be?

Every field has value, every field is keyword searchable, so it?s fine. It?s important that you?re not including more information than it?s really needed. It?s not about the length.

8 ) How do you recommend that job searchers ensure that they have online images that are positive instead of negative?

The key to stay positive and don?t complain, don?t be a negative nancy. ?Never make comments about your job, your boss, how bored you are at work, how frustrated you are that you?ve been in a job market for year and they won?t hire you yet. Posting a positive comment even if you don?t really feel it, it puts a positive thought in your head as well. Be active. Wherever you have sites that you want to show at the top of Google, that?s where you must have the most activity. Continue googling yourself to see what?s showing up on page one.

9) What are some common mistakes that you see job seekers making right now?

The generic resume is one of them. Being negative is another. But honestly the biggest one is giving up. Buying into the idea that you?re not worth it, you don?t have enough to offer. Letting that frustration, that is natural, affect your psyche because that will be requested in your interviews and interactions with employers. What I recommend is that job seekers should go back re-read their resumes, re-read their old performance review, look at all the great things that they have done. Remind yourself that you matter and you made a difference to your employers and you will make the difference for the next one just because it hasn?t happened yet it doesn?t mean it isn?t going to soon.

10) This question I ask at the end of all of my interviews, what do you wish you would have known when you were a young professional woman just starting out in her career?

Be patient. It?s easy to think I have finished my degree and I have so much to offer. Yes, you might be really bright and have a lot to offer but it?s the beginning of your career and you have a lot to learn. Be patient and recognize that this thing will happen. I don?t mean to sit back and wait. Just because you are not selected for that promotion then that?s the sign that it will never happen and you should leave that company. ?That is a really important big decision and it?s important to get some feedback from someone that you know as a mentor who knows what?s going in the professional world. Find out what are the things that you need to develop.

Calif. blackouts possible with nuke plant offline

(AP) ? Southern California utility officials are warning that blackouts in the region are possible this summer as a result of the sidelined San Onofre nuclear power plant.

The damaged plant is likely to remain sidelined until at least the end of August while investigators probe excessive wear in tubing that carries radioactive water, the plant's operator said Thursday.

The officials say that if a heat wave hits while the twin-reactor plant is offline, rotating blackouts are a possibility. Utilities have been scrambling to find replacement power as a precaution, including restarting two retired natural gas-fired plants in Orange County.

Southern California Edison said in a statement that the company plans by the end of July to submit a plan to federal regulators to restart the Unit 2 reactor, where damage to tubes in its steam generators has been less severe than in its twin, Unit 3.

A proposal to restart either reactor must be approved by the Nuclear Regulatory Commission, and that review could take weeks or longer.

Edison spokeswoman Jennifer Manfre said it's likely the plant between San Diego and Los Angeles will remain shuttered at least through August.

The trouble began to unfold in January, when the Unit 3 reactor was shut down as a precaution after a tube break. Traces of radiation escaped at the time, but officials said there was no danger to workers or neighbors. Unit 2 had been taken offline earlier that month for maintenance, but investigators later found unexpected wear on hundreds of tubes in both units.

Gradual wear is common in such tubing, but the rate of erosion in some tubes at San Onofre alarmed officials since the generators are relatively new. The company has said 1,300 tubes will be taken out of service, although the number is well within the margin to allow the generators to keep operating.

The company has found that the tube wear is being caused by vibration and friction with adjacent tubes and bracing, but investigators have yet to say why that's happening or how they will fix it.

Edison initially targeted a June restart for at least one of the twin reactors. A tentative plan called for restarting and running the seaside reactors at lower power, at least for several months, because engineers believe that will ease vibration that could be eroding tubes.

The NRC has said there is no timetable to restart the reactors, which were replaced in 2009 and 2010 in a $670 million overhaul.

About 7.4 million Californians live within 50 miles of San Onofre, which can power 1.4 million homes.

Safety issues at the plant have attracted congressional scrutiny, and some officials in nearby communities have been calling for San Onofre to shut down permanently. The Irvine City Council urged the NRC to thoroughly review safety conditions at the plant before it is considered for relicensing in 2022.

Edison has said safety for the public and its employees is the company's top concern.

The plant is owned by SCE, San Diego Gas & Electric and the city of Riverside. The Unit 1 reactor operated from 1968 to 1992, when it was shut down and dismantled.

Scientists find new role for P53 genetic mutation -- initiation of prostate cancer

A team of UC Davis investigators has found that a genetic mutation may play an important role in the development of prostate cancer. The mutation of the so-called p53 (or Tp53) gene was previously implicated in late disease progression, but until now has never been shown to act as an initiating factor. The findings may open new avenues for diagnosing and treating the disease.

The study was published online in the journal Disease Models & Mechanisms and will appear in the November 2012 print edition in an article titled, "Initiation of prostate cancer in mice by Tp53R270H: Evidence for an alternate molecular progression," and is available online.

"Our team found a molecular pathway to prostate cancer that differs from the current conventional wisdom of how the disease develops," said Alexander Borowsky, associate professor of pathology and laboratory medicine and principal investigator of the study. "With this new understanding, research can go in new directions to possibly develop new diagnostics and refine therapy."

Prostate cancer is the leading cancer diagnosis in men in the United States. Although it is curable in about 80 percent of men with localized disease, the rate is much lower if the cancer is highly virulent and has spread beyond the prostate gland.

The investigators developed a mouse model genetically engineered to have a mutation in the "tumor suppressor" gene, p53, specifically in the cells of the prostate gland. These mice were significantly more likely to develop prostate cancer than control mice without the mutation, and provided the first indication that the p53 mutation could be involved in the initiation of prostate cancer. They also note that the mutation of p53 in the prostate differs from loss or "knock-out" of the gene, which suggests that the mechanism is more complicated than simply a "loss of tumor suppression" and appears to involve an actively oncogenic function of the mutant gene.

The p53 gene encodes for a protein that normally acts as a tumor suppressor, preventing the replication of cells that have suffered DNA damage. Mutation of the gene, which can occur through chemicals, radiation or viruses, causes cells to undergo uncontrolled cell division. The p53 mutation has been implicated in the initiation of other malignancies, including breast, lung and esophageal cancers.

Other studies have associated p53 mutation with disease progression in prostate cancer, but this is the first to find that it can have a role in the early initiation of prostate cancer, as well.

Until now, understanding of the role of p53 was that mutation occurred exclusively as a late event in the course of prostate cancer. Based on the findings in the new mouse model that the researchers developed, p53 mutation not only can initiate prostate cancer but might also be associated with early progression toward more aggressive forms of the disease.

Genetic mutations can initiate cancers in a variety of ways. Those include promotion of uncontrolled cell growth and loss of the gene's normal cell growth-suppressor functions. Exactly how the p53 mutation promotes the initiation and progression of prostate cancer remains to be clarified and is a focus of current research by the UC Davis team. They also are trying to gain an understanding of how the p53 mutation affects the effectiveness of standard treatments for prostate cancer, such as radiation and hormone therapy.

Another application of the discovery could be the development of a new diagnostic test for prostate cancer based on the presence of the p53 mutation as a biomarker.

"Knowing that prostate cancer can develop via p53 mutation opens new opportunities for researchers in the field," said Borowsky. "This is a game-changer in the understanding of prostate cancer."
