Light-induced delivery of nitric oxide eradicates drug-resistant bacteria

Researchers at the University of California, Santa Cruz, have developed a novel approach for eradicating drug-resistant bacteria from wounds and skin infections, using light to trigger the controlled release of nitric oxide. The UCSC team developed a photoactive compound that releases nitric oxide when exposed to light, and loaded it into a porous, biocompatible material that could be applied as a sprayable powder.

In laboratory tests, the light-triggered nitric oxide treatment eradicated a highly drug-resistant strain of Acinetobacter baumannii, a type of gram-negative bacteria that causes hard-to-treat and potentially lethal infections throughout the world, including serious infections in soldiers wounded in Iraq and Afghanistan. The team led by Pradip Mascharak, professor of chemistry and biochemistry at UC Santa Cruz, and graduate student Brandon Heilman published their results in the Journal of the American Chemical Society (JACS). The paper is currently available online and will be featured on the cover of a future print issue of the journal.

Nitric oxide has potent antimicrobial effects and is known to play a role in the immune system and promote wound healing. Gaseous nitric oxide has been used to treat infected wounds, but handling the toxic and reactive gas poses many challenges. So researchers have begun exploring a variety of other methods for delivering nitric oxide as an antibiotic treatment. Because nitric oxide attacks a large number of targets in microorganisms, including DNA, proteins, and lipids, many scientists expect bacteria will not easily develop resistance to it.

Mascharak's lab developed a photoactive manganese nitrosyl, a compound that rapidly releases nitric oxide when exposed to light. As a carrier for this compound, the researchers used a porous silicate material known as MCM-41, which traps the photoactive compound inside its pores. They also tested a related aluminosilicate material (Al-MCM-41), which holds the photoactive compound even more tightly. Tests showed that after the light-triggered release of nitric oxide, the byproduct of the reaction remains trapped inside the powdery, biocompatible material.

"It only delivers nitric oxide. The rest remains trapped in the material, which can be washed out of the wound," Mascharak said. "We think it could be used as a sprayable powder for treating battlefield wounds."

Acinetobacter baumannii has earned the nickname "Iraqibacter" because it has caused so many serious infections in soldiers wounded in Iraq. Some strains of the bacteria are resistant to virtually all antibiotics. Mascharak's lab tested their compound against a strain, isolated from a soldier injured in Afghanistan, that showed resistance to nine of 11 antibiotics tested.

To test the photoactive compound, the researchers developed a laboratory model of skin and soft-tissue infections. A standard antibacterial assay involves growing bacteria on the surface of an agar plate (a petri dish with a layer of firm, gelatin-like growth medium). In an infection, however, bacteria are not only on the surface but also deeper within the skin or soft tissues. "We realized that there wasn't a good model for in vitro testing of antibiotics against soft-tissue infections," Heilman said.

To more closely mimic the conditions in an infected wound, Heilman mixed bacteria into a warm solution of "soft brine agar" and poured that onto agar plates to solidify. The bacteria then grew throughout a 1.1-millimeter-thick layer of soft agar, allowing growth and colonization to occur in a manner similar to that seen in skin and soft-tissue infections.

Heilman then applied the aluminosilicate powder, with and without the photoactive manganese nitrosyl compound, to a defined area of the plates before shining visible light on them. The released nitric oxide effectively cleared the bacteria from the treated areas of the plates, showing that the nitric oxide easily penetrated through the agar layer. The amount of light used to activate the compound (100 milliWatts per square centimeter) is a typical light flux on a sunny day, Mascharak said.

The ability to control the release of nitric oxide using light is a significant advantage for clinical applications, he added. Tests showed that illumination of the material causes a steady release of nitric oxide, which can be stopped and started repeatedly by turning the light off and on. In the field, this could be accomplished by covering and uncovering the treated area.

"This is the first proof-of-concept to show that it works," Mascharak said. In the paper, the researchers concluded that "It is expected that application of these nitrosyl-containing porous materials to infected wounds followed by exposure to sunlight will bring about a rapid reduction of the pathogen load."

The researchers now hope to find collaborators who can help them with the next levels of testing needed to develop the clinical potential of their compound.

A little over a year ago, I wrote about the importance of good health and good physical fitness to long term survival.? After huffing and puffing during a recent hike, I realized that the time had come to reassess my personal health and fitness goals and to bone up on personal medical preparedness.

Somewhat egging me on was a recent reminder from Dr. Bones (aka Dr. Bones at the Doom and Bloom Nation and co-author with Nurse Amy of The Doom and Bloom Survival Medicine Handbook), that this is one area of survival prepping that tends to fall to the bottom of the priority list.

But surely, we are remiss if we overlook the positive aspects good health and both physical and mental fitness during times of stress. I don?t know about you, but for me the ultimate stress would be a major natural or man-made catastrophe.

Think about it. You might have six months of food, six months of water, a sustainable food garden, a fully stocked first aid kit, tools, supplies and generators that would allow you to live off the grid if the SHTF.? But what if you had to flee? What if you had to grab your boots, bags and backpacks and really get the heck out of dodge? Could you make it? How far could you walk in dangerous weather conditions or uncertain terrain? And the stress. Could you cope?

The Personal Fitness Inventory

These are all important questions that might be answered by taking a personal health and fitness inventory by asking the following questions:

Health:? What the state of your general health?? Do you take a lot of prescription drugs?? Is your weight in proportion to your height?? Do you check your blood pressure regularly and do you see a dentist at least once a year for a general checkup and cleaning?

Fitness:? Do you exercise regularly?? How far can you walk briskly without getting winded?

Mental Balance:? Are you happy?? Can you cope with the stresses of daily life?? Do you consider yourself mentally ?balanced??

It is my belief that these three survival tools (health, fitness and mental balance) work together so that improvement in one area should help the other two.? Want some recommendations?

1. If you are overweight or have a curable health condition, what are you waiting for? Now is the time to start that diet, fix that bum knee, and yes, get those teeth taken care of. Stock up on some supplements (a multivitamin, calcium, magnesium, fish oil) and don?t forget to take them. Even if you don?t have health insurance, shop around for some low cost clinics and make the investment in your health.
2. I believe that the best piece of fitness equipment is what nature gave us, our own two feet. That means that with comfortable clothes, some hiking shoes or sneakers and the will to move, we can get in shape pronto. All that is required is a bit of time, some place to go, and if you are so inclined, an mp3 player with music or an audio book.
3. Mental balance is a toughie because what works for one, may not work for someone else. The Mayo Clinic says that the effects of stress on the body include:

Evaluate the stresses in your life and work on the one or two items over which you have some control.? For example, if the clutter in your home is driving you nuts, take an hour a day for the next week to de-clutter your living space.? When that it done, tackle the next item.? The key is to set aside and forget about those items you cannot control ? you will only frustrate yourself and set yourself up for failure.

It always surprises me how great a difference even the smallest change can make to one?s well-being.

Without being overly simplistic, working on these three aspects of health preparedness will go a long way toward seeing us through a disaster, a personal or economic crises or even when the SHTF.? But there is more to it than simply good health.?

Medical Preparedness ? An Open Letter to Preppers

These minor issues, so easily treated by modern medical science, can easily become life-threatening if left untreated in a collapse scenario.

Don?t you owe it to yourself and your family to devote some time and effort to obtain medical knowledge and supplies?? The difficulties involved in a grid-down situation will surely put you at risk for sickness or injury. It?s important to seek education so that you can treat infectious disease and the other ailments that we?ll see.? There will likely be a lot more diarrheal disease than gunfights at the OK corral.? History teaches us that, in the Civil War, there were a lot more deaths from dysentery than there were from bullets.?

Some say 'Beans, Bullets and Band-Aids', but I say 'Beans and Band-Aids, then Bullets'.? I suppose, as a physician, that?s not too surprising.

If you make the commitment to learn how to treat medical issues and to store medical supplies, you?re taking a genuine first step towards assuring your family?s survival in dark times.? The medical supplies will always be there if the unforeseen happens, and the knowledge you gain will be there for the rest of your life.? Many medical supplies have long shelf lives; Their longevity will one of the factors that will give you confidence when moving forward.

It?s important to know some illnesses will be difficult to treat if modern medical facilities aren?t available.? It will be hard to do much about those clogged coronary arteries; there won?t be many cardiac bypasses performed.? However, by eating healthily and getting good nutrition, you will give yourself the best chance to minimize some major medical issues.? In a collapse situation, an ounce of prevention is worth, not a pound, but a ton of cure.? Start off healthy and you?ll have the best chance to stay healthy.

When I say to obtain medical knowledge, I am also encouraging you to learn about natural remedies and alternative therapies that may have some benefit for your particular medical problem.? Essential oils, herbal remedies, and other time-honored methods of healing should not be overlooked as you put together your medical supplies.? I cannot vouch for the effectiveness of every claim that one thing or another will cure what ails you; suffice it to say,? that our family has an extensive medicinal garden and that it might be a good idea for your family to have one, also.? Many herbs that have medicinal properties grow like weeds, so a green thumb is not required to grow them.? Many of them do not even require full sun to thrive.? Even the White House garden has a collection of these beneficial plants.

I?m not asking you to do anything that your great-grandparents didn?t do as part of their strategy to succeed in life.? In a collapse situation we?ll be thrown back, in a way, to that era.? We should learn some lessons from the methods they used to stay healthy.? Those homespun skills will give you a head start to being an effective medic for your family.

The non-prepper members of my family wonder why I spend all my time trying to prepare people medically for a collapse situation.? They tell me that I can?t turn everyone into doctors, so why I should try??? Am I trying to turn you all into doctors?? No, there?s too much to learn in one lifetime; even as a physician, I often come across things I?m not sure about.? That?s what medical books are for, so make sure that you put together a library. You can refer to them when you need to, just as I do.?

I AM trying to turn you into something, however: I?m trying to make you a better medical asset to your family and/or survival community than you were before.? If you can learn how to treat common illnesses and injuries in times of trouble, I will have succeeded in my mission.

CNET is reporting today that the FBI has quietly created a new web surveillance unit, charged with the task of developing new ways to intercept online, wireless, and VoIP communications. The Bureau has thus far been rather tight-lipped about its unit, known as the Domestic Communications Assistance Center, declining to name even the person in charge of it. CNET, however, was able to glean and piece together information about the initiative through interviews and reviews of internal government documents.

According to these sources, DCAC is designed to cover a relatively broad scope of surveillance- and security-related issues. Its mandate includes intercepting Skype messages, building wiretap devices, and serving as a ?surveillance help desk? for local, state, and federal authorities. The outfit will also be responsible for analyzing any data a provider or social network may submit in response to court orders. A source close to the matter told CNET that the DCAC is currently being launched, though it?s not yet operational. A Justice Department document, however, describes the agency as ?recently established.?

The FBI has long pushed for a more comprehensive approach to electronic surveillance, on the grounds that recent encryption developments have made it far more difficult for investigators to intercept communications. In February, then-general counsel Valerie Caproni testified before the House Judiciary Committee, arguing for new legislation that would require social networks and wireless providers to have clear procedures for divulging encrypted data when requested to do so by the government.

Valerie added that new laws were needed because ?individually tailored solutions have to be the exception and not the rule.? According to CNET?s sources, these ?individually tailored solutions? are exactly what DCAC has been charged with developing ? customized surveillance technologies designed for isolated companies or individuals.

Internal documents show that the DCAC has been in the works since at least 2008, when FBI director Robert Mueller received a briefing on it. Until recently, though, it?s remained shrouded in secrecy. The unit has no website, has never been formally announced, and has only received passing mention in public budget requests.

This secrecy has raised concerns among some civil libertarians, including Jennifer Lynch, an attorney at the Electronic Frontiers Foundation. ?We should know more about the program and what the FBI is doing,? Lynch told CNET. ?Which carriers they?re working with ? which carriers they?re having problems with. They?re doing the best they can to avoid being transparent.?

The FBI did provide a statement on its new center (referred to here, and elsewhere, as the National Domestic Communications Center, or NDAC), emphasizing that the unit will only provide the technical foundation for wiretap investigations, without actually performing any surveillance itself. ?It is important to point out that the NDCAC will not be responsible for the actual execution of any electronic surveillance court orders and will not have any direct operational or investigative role in investigations,? the statement reads. ?It will provide the technical knowledge and referrals in response to law enforcement?s requests for technical assistance.?

Sony Xperia U goes on sale at Three UK

We've been twiddling our thumbs waiting patiently for the littlest of Sony's new-look smartphone lineup, the Xperia U, to arrive in earnest. It's a bit of a relief then to see the Android 2.3 phone go on sale at Three UK. You'll be getting what we saw back in Barcelona, including Sony's Timescape interface and the trick color light-up strip. We're guessing that most will see the cheap-as-chips price as the real selling point: the Xperia U costs £170 ($269) without a plan, and it's free as in beer on a two-year contract for £23 ($36) per month. O2, Orange and T-Mobile will expand UK carrier choices soon, but if you're looking for a fresh entry point into Android, Three can take care of you today.

Bellagio robbery foiled: Thief loses wig in $155,000 attempted heist

Bellagio robbery that wasn't. Two thieves sprayed a blackjack dealer at the Bellagio casino in Las Vegas with mace and tried to steal $115,000 in casino chips. One was tackled and caught. The other escaped.

A hapless bandit lost his wig, sunglasses and $115,000 worth of casino chips when security wrestled him to the floor during a botched weekend heist at a posh Vegas Strip resort, authorities said Monday.

Two men attempted to rob the Bellagio on Saturday night by spraying a blackjack dealer and others with a caustic eye-burning chemical, police said.

With the distraction in the air, Michael Q. Belton snatched up nearly two dozen high-value chips and took off for the door, according to a police report.

The man who sprayed the noxious gas escaped, but Belton was tackled to the floor and held until police arrived.

Belton struggled at first, according to the police arrest report, but then suddenly stopped fighting back against casino employees and surrendered.

He dropped 23 red, white and blue chips, valued at $5,000 each, police said.

"How long am I going away for?" Belton asked detectives during a recorded interview following his arrest, according to the report.

Investigators said Belton, of southern California, told them he needed the money because he is unemployed and his grandparents are ill.

He told police he didn't know the man who got away. Bolton told investigators he responded to an Internet posting for a job repossessing cars. But he said when he reached Las Vegas, the man who posted the job ad said he wanted him to help another man rob the Bellagio.

Belton was held Monday on $60,000 bail at the Clark County jail pending an initial court appearance Tuesday on felony robbery, conspiracy and burglary charges. It was not immediately clear whether he had a lawyer.

Officer Jose Hernandez, a department spokesman, said the investigation was ongoing and Belton would not agree to a jail interview.

The suspect who got away apparently did not take any chips as he fled. Police said all chips were recovered, and casino officials say they sustained no financial loss.

Authorities said they were still searching for the second man in the heist, which they said was recorded by casino security cameras. The video was not made public.

No serious injuries were reported.

The last bandit to make off with casino chips from the Bellagio, in December 2010, was sentenced to three to 11 years in state prison for armed robbery and assault with a deadly weapon. Anthony Michael Carleo wore a motorcycle helmet as he waved a gun and made off with $1.5 million worth of chips. He was arrested trying to redeem a $25,000 chip.

He was sentenced to an additional six to 16 years for another armed robbery at another Las Vegas casino.

He blamed addictions to drugs and gambling.
